Iontas will be presenting two talks at this years PepTalk:
Dr John McCafferty (Iontas CEO) will be presenting: Generating Ion Channel Blocking Antibodies by Fusing Cysteine-Knot Miniproteins into Peripheral CDR Loops at 10.45 on Monday 8th January. The talk will describe the use of Cysteine-knot miniproteins (knottins) as therapeutic agents to block proteases and ion channels involved in cancer, autoimmunity and pain. The talk will highlight the manufacturing difficulties, short half-lives, and a lack of specificity of these agents and how functional knottins can be inserted into peripheral antibody CDRs via short linkers to potentially address these issues. The resulting “KnotbodyTM” that have been developed retain the blocking activity from the knottin while enjoying the extended half-life and additional specificity conferred by the antibody molecule.
Dr Mike Dyson (Iontas CTO) wull be presenting presenting: Mammalian Display: Antibody Discovery, Affinity Maturation and Developability Screening in IgG Format at 4.30 on Monday 8th January. The talk will describe the use of directed integration of antibody genes by CRISPR/Cas9 and TALE nucleases, to construct large libraries in mammalian cells containing a single antibody gene/cell. This permits construction of millions of monoclonal stable cell lines displaying IgG antibodies on their surface from which antibodies can be selected by flow cytometry for specificity, binding affinity, species cross-reactivity, developabilty and expression level.
