IONTAS uses the principles of targeted oligonucleotide-directed mutagenesis to achieve improved binding affinities and, if desired, cross reactivity with other animal species or related targets.
Our in vitro affinity maturation service typically performs three rounds of phage display to generate antibodies with enhanced binding. The technology has been successfully used to optimize antibodies and antibody fragments to create antibodies with sub-nanomolar binding affinities. In addition, chain shuffling can be used to increase the genetic diversity and the numbers of affinity matured antibodies.
With oligonucleotide-directed mutagenesis, the antibody variable regions, typically CDR3, are mutagenized by the targeted randomization of certain positions at a defined diversity. The resultant sub-libararies are then be selected to identify antibody variants with improved binding properties. Engineering of the antibody can improve affinity, potency in functional assays , introduce cross species binding and remove binding to related target family members . In doing so, sub-libraries are created which then undergo selective screening. Selected populations showing the desired binding properties are then expressed either in an scFv format or as IgG and subjected to affinity measurements using Surface Plasma Resonance and cell based functional screening.
